It varies by location. Some do with a weight limit and a deposit. Others theres just a deposit, and most do not allow pets at all.
It's really easy to tell if there is a pet in the room, or there has been a pet in the room! It's called Housekeeping! If the hotel/motel is pet free, then you can bet your paycheck someone will find out. Either Housekeeping or another guest will say something to the front desk.
Most Hilton properties do not accept pets. Although I believe the NY Hilton may, but you should verify this. I know the Comfort Inn accepts pets, but they have a weight limit and require a deposit. You may have to check with Marriott?

Yes, they care. They may not know, but if they find out, you can find a pet charge added to your bill.
CALL the hotel involved and find out what their policy is. Some do permit pets and sometimes there is an additional fee.
If you take a pet into a no pet hotel and are found out, they have every right to kick you out.
